4) Difficulty in definitive establishment of a candidate gene effect.

• Identifying the causal variant for a candidate gene effect may be possible for major genes such as the halothane sensitivity gene.
• In cases of minor quantitative effects, identification of the site differences associated with a given candidate gene effect may not be feasible.
